Devil's Claw Extract (Harpagophytum Procumbens Root Extract): Harpagoside as a Lead Marker for Antioxidant Formulations
Devil's Claw Extract (INCI: Harpagophytum Procumbens Root Extract) is obtained from the root of Harpagophytum procumbens and is available to formulators in three carrier variants: Glycerin, Propylene Glycol and Pentylene Glycol. The extract content is approximately 30% across all variants. As a plant-based, vegan raw material, the extract is suitable for cosmetic applications under EC No. 1223/2009 as well as, in the Glycerin variant, for functional food applications. This article summarises the markers documented in the data sheet, the efficacy profile and the technical parameters for formulation development. Harpagoside: the characteristic iridoid glycoside lead marker The lead marker of the extract is harpagoside, an iridoid glycoside. The product specifies an indicative content of 122 to 945 µg/mL. At a use concentration of 2%, this results in a calculated 2.4 to 18.9 µg/mL harpagoside in the finished formulation. For food and beverage applications, a dosage of 1.3% yields 1.5 to 11.8 µg/mL accordingly. In addition to harpagoside, further characteristic compounds of the drug have been identified: procumbide (0.1 to 0.3%) and harpagide (0.5 to 1%). These markers allow analytical characterisation of the root extract and support specification testing during incoming goods inspection. Documented efficacy profile and molecular targets The CDS lists antioxidant, anti-inflammatory and capillary-strengthening properties for Devil's Claw Extract. The cosmetic claims documented are "Balancing", "Elasticity support" and "Anti-inflammatory". At the molecular level, the targets identified are ROS scavenging, MMP inhibition and NF-κB. These statements refer to the cosmetic and food technology application spectrum of the raw material. They are not to be understood as medical or disease-related claims. The final evaluation of claims for a specific end product is the responsibility of the responsible person in accordance with the applicable regulatory requirements. Three carrier variants for different formulations The choice of carrier determines the field of application: Glycerin variant (INCI: Glycerin, Harpagophytum Procumbens Root Extract): anhydrous glycerin (≥ 99.5%) accounts for approximately 70% of the final product; purified water serves as the extraction solvent and does not remain in the final product. This variant is also suitable for food and functional food applications. Propylene Glycol variant (INCI: Aqua, Propylene Glycol, Harpagophytum Procumbens Root Extract): aqueous propylene glycol/water mixture. Pentylene Glycol variant (INCI: Aqua, Pentylene Glycol, Harpagophytum Procumbens Root Extract): indicative values approx. 50% water, approx. 30% extract, approx. 20% Pentylene Glycol. All variants are specified as clear liquids with a characteristic colour (amber to light brown) and are suitable for cosmetic applications. Use concentration and physical parameters The recommended use concentration in cosmetics is 2 to 5% (specified in the CDS as 1 to 3% as an efficacy-related range). For food and functional food applications, 0.5 to 2% is indicated. The relative density is between 1.03 and 1.06. Specific individual values for pH, density and refractive index should be taken from the product-specific CoA. Note on storage stability: depending on temperature, colour changes may occur during the storage period; in the event of turbidity or precipitation, filtration is possible without impairing product quality. The shelf life is 24 months when stored dry between 10 and 30 °C in the original sealed container. Safety and documentation According to the SDS, Devil's Claw Extract is not a hazardous substance within the meaning of Regulation (EC) No. 1272/2008. No hazard pictograms, signal word or H/P statements are assigned; PBT/vPvB and endocrine-disrupting properties (≥ 0.1%) are not present. The raw material is not classified as dangerous goods under ADR/RID, IMDG and IATA.
