Hibiscus Powder Extract 10:1 (Hibiscus Sabdariffa): Delphinidin-3-sambubioside as an Anthocyanin-Rich Lead Marker

The Hibiscus Powder Extract 10:1 (INCI: Hibiscus Sabdariffa) by CONIUNCTA® is obtained from the calyces of Roselle and is available as a spray-dried, concentrated powder. This extract is characterized by its content of anthocyanins, most notably Delphinidin-3-sambubioside. As an EU organic-certified raw material (DE-ÖKO-001), the powder is suitable for both cosmetic and food and dietary supplement applications. For formulators, the powder form offers advantages in terms of dosing accuracy, storage stability, and integration into powder- or tablet-based systems.

Delphinidin-3-sambubioside: the anthocyanin lead marker

The primary marker of this extract is Delphinidin-3-sambubioside, a member of the compound class of anthocyanins. According to the Claim Data Sheet, the indicative content in the extract is 70 to 270 mg/g. This range results from a literature content of 1 to 3% in the dried plant, the 10:1 concentration factor, and an assumed recovery of 70 to 90% (source: Da-Costa-Rocha et al. 2014). Anthocyanins are responsible for the characteristic coloring of Sabdariffa calyces and represent the documented lead marker in the extract.

With an extract usage of 2% in the formulation, the powder mathematically delivers 1.4 to 5.4 mg/g Delphinidin-3-sambubioside in the finished formulation. This figure allows formulators to transparently calculate the marker content in the final application.

Further documented compounds

In addition to the anthocyanin lead marker, the CDS lists two further compounds: chlorogenic acid at a content of 0.3 to 1% and hibiscus acid at 1 to 3%. Both belong to the characteristic phytochemical profile of Sabdariffa calyces and complement the marker spectrum of the extract. The CAS number of the extract (84775-96-2) is recorded for the composition.

Production as a spray-dried powder

The hibiscus powder extract is produced by spray-drying a 10:1 concentrated extract. Ethanol and water are used as extraction solvents. Maltodextrin serves as the carrier system at a proportion of 1 to 20%; accordingly, the proportion of the hibiscus extract is 80 to 99%. The result is a brown powder with a particle size of 80 mesh (at least 95% passage). According to product specifications, the bulk density is between 0.50 and 0.70. Appearance, odor, and taste are described as characteristic.

An important note for processing: due to the anthocyanin content, temperature-dependent color changes may occur during storage. This is inherent to the product and does not affect the specified parameters at the time of shipment. The specified color ranges from golden yellow to light brown.

Documented profile of effects and application areas

The claim area supported in the CDS is "Antioxidant protection", based on the phytochemical profile and published literature. The data sheet lists ROS scavenging and collagenase inhibition as cosmetic targets. For food applications, metabolic health and the reduction of oxidative stress are listed as documented application areas. We expressly point out that every claim requires independent substantiation by the finished product manufacturer in accordance with Regulation (EC) No 655/2013.

For cosmetics, the CDS states a general usage rate of 2 to 5% as well as a use level (claim table) of 1 to 3%. In functional foods and beverages, a use level of 0.5 to 2% is specified. The extract is suitable for vegan formulations and intended for use under Regulation (EC) No 1223/2009 (cosmetics) as well as in the food sector. Processing, storage and safety

As a powder extract, the raw material integrates into water-based systems as well as powder-form applications such as tablets, capsules, or instant beverage powders. Recommended storage is cool, dry, and well-ventilated in an airtight container, protected from heat and moisture; the SDS specifies a storage temperature of 15 to 25 °C. When stored dry in the original sealed container, the shelf life is 24 months. In terms of safety, the product is not classified as a hazardous substance under Regulation (EC) No 1272/2008; no H- or P-statements are required. Respiratory protection is recommended in case of aerosol formation, and safety goggles are also advised. The product is not classified as dangerous goods under ADR/RID, IMDG, or IATA.

The hibiscus powder extract is EU organic-certified under Regulation (EU) 2018/848 with certificate number DE-ÖKO-001 (Kiwa BCS).
